Of course you can use Windows Media Player to delete music on your Sansa.  I do it all the time. 
 
I'm going to describe WMP 10 -- it may be a littlle different if you have WMP 11, but you'll figure it out. 
 
1)  Plug in your Sansa (in MTP mode, if you switched it to MSC; don't worrry about it if you didn't switch it). 
 
2)  Open WMP. 
 
3)  Go to the "Sync" tab -- the same one you use to put music onto your Sansa. 
 
4)  The box on the right hand side of the screen shows all the music in your player.  Click your way down to the music you want to delete.  Select the music, and hit the "delete" button on your keyboard.  Gone! 
 
 
Post here if you have any problems with this.

Where do you switch it from MSC to MTP mode?

I don't have the E250, however I am guessing it might be like later Sandisk players, wheere the USB mode is set through settings, system settings, USB mode.
